Although these monoamine-based agents are potent antidepressants, the cause of depression is far from being a simple deficiency of central monoamines. The glutamatergic system also plays important roles in the pathophysiology of major depressive disorder. This paper summarizes the physiological characteristics of glutamate in the brain, glutamatergic abnormalities in depression andantidepressants acting on the glutamatergic system.
